Name of the mutant parasite
RMgm numberRMgm-5403
Principal nameΔPbMAP1
Alternative name
Standardized name
Is the mutant parasite cloned after genetic modificationYes
Phenotype
Asexual blood stageReduced growth/multiplication of blood stages in mice (mice infected with ΔPbMAP1 parasites died 4–10 days later than those infected with wild type).
Gametocyte/GameteNot tested
Fertilization and ookineteNot tested
OocystNot tested
SporozoiteNot tested
Liver stageNot tested
Additional remarks phenotype

Mutant/mutation
The mutant lacks expression of PbMAP1

Protein (function)
A protein of 1303 amino acids, which is predicted to contain a signal peptide at the N-terminus. In the paper evidence is presented that it binds heparin and to mouse erythrocytes and is located in merozoites (on the plasma membrane).

Phenotype
Reduced growth/multiplication of blood stages in mice (mice infected with ΔPbMAP1 parasites died 4–10 days later than those infected with wild type).

Promoter of the selectable markerpbdhfr
Selection (positive) procedurepyrimethamine
Selection (negative) procedureNo
Additional remarks genetic modificationThe plasmid was constructed using pl0001 (MR4; ATCC,
USA). PbMAP1 5′- and 3′-untranslated regions (UTRs)
were PCR-amplifed as the left or right homologous arms
and inserted respectively upstream and downstream of
the DHFR gene. Te 5′-UTR and 3′-UTR were subcloned
in the pl0001 plasmid using restriction enzyme sites (Apa I/BspD I or BamH I/Not I). The plasmid was digested with Kpn I and Sac II before transfection.
