Mutant/mutation
The mutant lacks expression of MDR5 and expresses the fusion gene of GFP and Luciferase under the control of the constitutive eef1a promoter

Protein (function)
In different Plasmodium parasites 14-16 ABC-transporter genes have been identified (16 in P. falciparum, 14 in P. berghei). Based on phylogenetic analysis of the conserved nucleotide-binding domains, seven of these are recognized as members of the B family of ABC transporters (both in P. falciparum and in P. berghei). This includes MDR5.

Phenotype
A reduced growth rate of asexual blood stages (mean multiplication rate of 6.8x per 24 hour in the cloning assay; wild type parasites show a multiplication rate of 10x per 24 hour in this assay). Normal gametocyte and ookinete production. Reduced numbers of (mature) oocysts. Strongly reduced numbers (>10 times) of salivary gland sporozoites. However, sporozoites show normal infectivity to mice. Intravenous injection of 10.000 mutant sporozoites resulted in blood infections in mice (n=3) with a prepatent period (5-6 days) that is comparable to the prepatent period of wild type parasites.

Additional information
Northern analysis and RNAseq analyses indicate that mdr5 is transcribed in blood stages. Northern analysis of blood stages of PbΔmdr5 demonstrated the absence of (correctly sized) mdr5 transcripts.
