The mutant expresses GFP under the constitutive hsp70 promoter

Phenotype
GFP expression of this mutant has not been described in detail for the different life cycle stages.
Ookinetes express GFP and show strong GFP-fluorescence. This mutant has been used to analyse motility and traversal behavior of ookinetes (see RMgm-168)

Additional information
This mutant has been crossed (in mosquitoes) with mutant RMgm-165, which lacks expression of CDPK3. A progeny mutant line from this crossing (RMgm-168), lacking expression of CDPK3 and expressing GFP has been used to analyse ookinete motility and traversal.

Additional remarks genetic modificationTo make a GFP expression cassette driven by the hsp70 promoter, the 1.2 kb upstream region and 1.0 kb downstream region of hsp70 were ligated to either side of the GFP coding sequence. To make a targeted insertion vector, this expression cassette was ligated into the downstream region of the pyrimethamine-resistant DHFR-TS gene. A transgenic GFP-expressing parasite clone (named WT:GFP) was obtained by inserting this construct downstream of the dhfr/ts locus by homologous recombination.
