The mutant lacks expression of UROD and expresses GFP under control of the constitutive eef1a promoter.

Additional information
The successful disruption of the gene encoding UROD shows that UROD is not essential for asexual blood stage growth/multiplication.

Evidence is presented that four genes in the de novo heme synthesis pathway PBANKA_1459200 (RMgm-1576), PBANKA_0608000 (RMgm-1577), PBANKA_1140700 (RMgm-1578) and  PBANKA_0105900 (RMgm-1579) can be disrupted without complementation.

Malaria parasites possess a de novo heme synthetic pathway, which is dispensable during  blood stage development in the mammalian host. The assembly of the two most important hemeproteins, cytochromes c and c1, is mediated by cytochrome heme lyase enzymes. Plasmodium spp. possess two cytochrome heme lyases encoded by separate genes. In P. berghei these genes are PBANKA_143950 and PBANKA_0602600. The unsuccessful attempts to disrupt these genes indicate an essential function during asexual blood stage growth/multiplication (see RMgm-1574 and RMgm-1575).
