Mutant/mutation
The mutant expresses a C-terminal HA-tagged version of PKG and expresses GFP under control of the constitutive eef1a promoter.

Phenotype
Mutant parasites show normal development throughout the complete life cycle. PKG-HA distributed throughout the cytoplasm of sporozoites and in liver stages (other stages not analysed).

Additional information
The mutant was used to determine the role of PKG in sporozoites and invasion of liver cells. Evidence is presented that PKG is required for sporozoite invasion of hepatocytes. Evidence is presented that that PKG is required for sporozoite motility, that PKG regulates the secretion of TRAP, an adhesin that is essential for motility and that PKG is involved in merozoite egress from infected hepatocytes.

See also mutant RMgm-1509: In this mutant the wild type pkg gene is replaced with a modified allele, pkg(T619Q-HA), in which the threonine gatekeeper residue was mutated to a larger glutamine residue together with a C-terminal triple HA epitope tag.
